Field et al. (2007) identified truncating mutations in the BRWD3 gene (Xq21.1) in 2 of 250 probands in families affected with XLID. These mutations were not identified in 520 control X chromosomes. In addition to mild to moderate ID, affected individuals in both families had macrocephaly, which was identified in infancy, prominent ears, and mild to moderate developmental delay. Language skills were the most severely delayed.

- Deletion/duplication analysis of the BRWD3 gene by CGH array is available for those individuals in whom sequence analysis is negative.
- Custom diagnostic mutation analysis (KM) is available to family members if mutations are identified by targeted mutation testing or sequencing analysis.
